Dimethylglycine, also known as DMG, is a powerful methyl donor with a uniquely fascinating history. Dimethylglycine was a part of the original vitamin B15 structure known as pangamic acid. Research over the years gave evidence that the biological activity ascribed to pangamate B15, was in fact, due to DMG.
DMG is an active amino acid and intermediate metabolite in the one-carbon Choline cycle. Its primary function is to contribute methyl groups for the methylation reactions and provide other essential building blocks. Scientists have been aware of DMG’s potential benefits for over a half-century, and it’s been used to enhance athletic performance for decades.* Its promise was first put to use as an addition to equine diets for racehorses in the 1970s and 1980s. Even though it has been used for decades, this phenomenal food substance’s benefits have only recently reached mainstream awareness in the general public.

DMG is a glycine-derivative amino acid classified as a food substance, not a mineral or vitamin. Even though it is found in foods such as beans and organ meat (like liver), it only exists in trace amounts.
The human body can synthesize significant amounts of dimethylglycine by metabolizing choline or losing one trimethylglycine methyl group through the homocysteine pathway. However, DMG is broken down so quickly and used in so many different pathways that its levels are spread thin and never get a chance to build up. As such, the most effective way of increasing quantities of dimethylglycine in the human body is through supplementation.
Because DMG is such a potent metabolic enhancer, it supports a plethora of systems in the human body.* It’s similar to the amino acid, glycine,
but with two methyl groups attached. The results of this 1981 double-blind placebo-controlled study with 20 human volunteers showed that DMG enhances humoral and cell-mediated immune response in humans.*
The researchers presumed DMG had potential as an immunomodulator because of studies done on calcium pangamate.* The study proved that the test group taking DMG had a greater than fourfold increase in antibody response to the pneumococcal vaccine as compared to the control group.* The DMG group also had a significantly higher overall response of leukocyte streptokinase-streptodornase, showing that it enhances the cell-mediated response as well.*
Researchers conducted a study in 2015 to investigate N, N Dimethylglycine, and its association with insulin resistance and metabolic traits using mass spectrometry-based metabolic approaches.*
Results showed consistent data points to the possibility of DMG deficiency playing a causal role in insulin resistance development. This study sparks the need for further studies to examine the inhibition of DMG-dehydrogenase (possible protective effects of DMG on glucose metabolism DMGDH-catalyzed reaction) and points to possibilities that DMG supplementation may be useful to maintain a normal insulin response.*
In 1990, researchers studied the impact of DMG on the immune response in New Zealand white rabbits. A group of rabbits were fed DMG for 14 days leading up to being exposed to immunocompromising factors.
The animals given DMG demonstrated a fourfold increase in mean antibody titer compared to controls.* Animals that were given DMG showed a tenfold increase in mean lymphocyte proliferative response (p = 0.0024) and a fourfold increase of lymphocyte production ( p = 0.0180).* This trial confirmed the results of the Grabor study that DMG can enhance both the humoral and cell-mediated response. No adverse effects or toxicity was observed at any time during the trial.
In 2016, researchers tested the cellular protective capabilities of DMG using early-development stage bovine embryos produced in vitro, which are known to be sensitive to reactive oxygen species.
After in vitro fertilization, presumptive zygotes were cultured with or without 0.1 μM DMG under different oxygen tensions. Findings proved that DMG enhanced the in vitro development of IVP bovine embryos by acting as a free-radical fighter.*
In this 2016 study, researchers compared the free-radical fighting abilities of DMG-Na and commonly used antioxidants, Trolox (6-hydroxy-2, 5, 7, 8-tetramethylchroman-2-carboxylic acid) against ABTS, DPPH, and H2O2.*
The goal was to test to see if lipid peroxidation’s contribution to the development of oxygen radical-related cell membrane damage could be reduced through enhanced antioxidant efforts. The free-radical scavenging activities of DMG proved to be the highest at 40mg/ml.*
Gastric intubation in mice with 12 mg DMG-Na/0.3 ml sterile saline solution significantly increased (P < 0.05) the body weight (BW) (28 d), organ proportion (liver and spleen), and antioxidant capacity in serum and the liver (Superoxide dismutase (SOD), Hydrogen peroxidase (CAT), Glutathione peroxidase (GPx), and Total antioxidant capacity (T-AOC).* These results indicate that DMG-Na could protect against the LPS-induced oxidative stress by enhancing the free radical scavenging capacity and increasing the activity of antioxidant defense systems.*
This study demonstrated that when DMG is made part of an equine’s diet, it can effectively reduce the build-up of lactic acid in the horses showing how DMG can improve their racing performance.* The horses on DMG were more aggressive, have better appetites and attitudes, and recover faster from racing and training than the control group of horses that did not receive the DMG.*
